Direct ELISA: In direct ELISA, antigens are immobilized in multi-well plates and detected by special conjugated primary antibodies.

Indirect ELISA: In indirect ELISA, the antigen is immobilized in multi-well plates and is first detected by unconjugated specific primary antibodies. An enzyme-conjugated secondary antibody is then administered against the host species of the primary antibody.

Sandwich ELISA: In sandwich ELISA, the capture antibody is immobilized in a multi-well plate. The sample is then added, followed by the detecting antibody. Capture and detection antibodies are specific for single and non-overlapping antigenic epitopes.

Both direct and indirect methods can be used to perform sandwich ELISA. Sandwich ELISA is used to accurately detect antigen concentrations in unknown samples and when high sensitivity and specificity of detection are required.

Competitive ELISA (also called inhibitory ELISA): In competitive ELISA, the antigen is immobilized in a multi-well plate and incubated with an antibody that has been previously incubated with the desired sample (which must also contain this specific antigen).
